In the Department of Chemistry at TU Darmstadt a position in the group of Prof. Dr. Katja Schmitz, Biological Chemistry, for a


Research Assistant (all genders) - 50 %:

is available in a temporary employment contract for an initial period of 3 years starting at the earliest possible date.

The advertised position involves
Rhodamine-labeled peptides and peptoids as biosensorsand substrates for enzyme-based assays.

In our research, we have found that cyclic peptides and peptoids labeled with the fluorescentdye tetramethylrhodamine fluoresce only slightly. However, when bound to a protein, this fluorescence is significantly enhanced.

In order to make this effect useful for biosensors and diagnostics, some questions need to be clarified:
How large must or may the macrocycles be? Which functional groups favor the effect? And with which other dyes can it be observed?


To clarify this, peptides and peptidomimetics will be synthesized on the solid phase, cyclized if necessary, and labeled with one or two fluorophores within the scope of the advertised position.

The purified peptides will be characterized spectroscopically and the change influorescence properties upon protein binding or cleavage of the peptide by proteases will be studied.

Based on these studies, improved biosensors and substrates for enzyme-based assays will be developed.


You have completed a bachelor's and master's degree in chemistry or biochemistry and have worked ideally on the synthesis and purification of peptides and peptide analogs, cyclization of peptides, fluorescence spectroscopy, fluorescence-based binding assays, or enzyme kinetic measurements as part of your specialization.

Relevant experience (e.g., in the context of internships or theses) with several of the above techniques is essential for the expeditious completion of the project.

Teaching experience, especially in supervising hall practicals, is desirable.
