Your Repsonsibilites

Build up and establish target validation workflow, using 2D patient derived cellular models (PDCM)
in knockdown/knockout experiments to assess typical readout parameters for (cancer) cell culture.

Generate 2D patient derived cellular models (PDCM) from cryopreserved tissues
Cell culture of 2D PDCMs
Establish and perform gene knockdown using siRNA/shRNA in 2D PDCM
Establishing of lentiviral vectors for transduction (mainly shRNAs)
Establish and perform gene knockout using CRISPR/Cas
Perform readout assays for typical cellular parameters in the context of cancer, e.g. viability, growth, migration etc.

Our Requirements

PhD in molecular biology, cell biology, biochemistry or similar
3 years working experience in transient and stable modification of gene expression in primary cells and functional screens, ideally in the context of drug target/biomarker validation or preclinical drug development
Strong background in patient derived cells or primary cells
Strong background in the abovementioned techniques
Experience with 3D PDCMs (organoids) is a plus
Experience with advanced cellular assays (e.g. coculturing) is a plus
Basic skills in MySQL, R etc. are desirable
Good language skills in English, German is a plus
