Your role
You will work closely with the Finn and Wilmes teams to aid in the selection of candidate virulence factors. You will assess the feasibility of recombinant protein production in different host systems and perform the expression and purification of the virulence factors of interest. Whilst developing the protein production workflow, you will interact regularly with the Letellier team to assure suitability and compatibility of the purified virulence factors with the planned downstream validation experiments. You will make use of different biophysical techniques such as for example nano-DSF, SEC-MALS, mass photometry and CD spectroscopy to characterize the purified virulence factors in vitro. When appropriate, experimental elucidation of the 3D structures of the newly identified virulence factors might be considered.

The minimum requirement for this position is a PhD degree in biochemistry, molecular biology or in a closely related scientific area. Expertise in the field of recombinant protein production in the milligram scale using multiple gene expression systems (E. coli, Pichia pastoris, insect cells) is crucial. A strong understanding and background knowledge of chromatographic techniques and extensive hands-on experience with the use of automated chromatography platforms for protein purification is absolutely essential. Experience with various biophysical characterisation methods is required. Teamwork, flexibility, strong organizational skills and the ability to interact with a multicultural community from different scientific backgrounds are expected. Fluency in English with excellent oral and written communication skills is necessary.
You might also have
Prior experience with the production of toxic proteins and virulence factors would be highly beneficial. Expertise in the field of protein production in Bacillus subtilis, other non-classical bacterial expression systems and/or cell-free expression methods would be considered a strong advantage for this position. A good working knowledge of various cloning methods would be very useful. Previous working experience with projects that involve the translation of dry-lab results to wet-lab experiments would be an important asset.
