- The project is based on published work by Dr. Julius Enssle, which describes the vaccination re-sponse and associated immunophenotypes of pa-tients with Multiple Myeloma during the SARS-CoV-2 pandemic. We established a framework to investigate humoral and cellular immune responses after immunization or infection at granular scale. Main findings from this work highlighted a reduced immune response in patients with Multiple Myeloma with striking differences between the B and T cell compartment. Multi-modal single cell sequencing revealed a relevant association of CD4+ cytotoxic T cells and activated and mature NK-cell populations with SARS-CoV-2 infection and/or SARS-CoV-2 vaccination. These results were published in the highly ranked scientific journals Blood, Leukemia and Cancer Cell (PMID , , ).
- The aim of the proposed project is to elucidate the immune cell phenotypes and alterations following SARS-CoV-2 and Influenza A/B vaccinations in Multiple Myeloma (MM) patients. In addition to the characterization of the humoral and cellular (non-) response, we focus on immune effector cell subsets and their transcriptional programs which govern their fate. Thereby, we aim to identify mechanisms and predictive models of MM patient-specific immunization responses.
- You will be tasked with planning, establishing and critically performing primary patient material measurements. Further, your work includes the computational analysis and modelling of the generated data and conducting follow-up in vitro experiments.
- To answer our scientific questions we are applying the following experimental techniques (among others): Serological immunoassays, plaque reduction neutralization tests, multi-color flow cytometry, flow-cytometry-based cell sorting, multi-modal single cell sequencing, BCR/TCR sequencing, bulk RNA sequencing, quantitative mass spectrometry-based proteomics, integrative bioinformatic and machine-learning-based data analysis and modeling.
- The applicant should hold a master degree or equivalent in biology, biochemistry, molecular medicine or related fields.
- A high level of motivation, flexibility and responsibility as well as very good communication skills in German or English are expected.
- Expertise in experimental techniques (e.g. high throughput sequencing analyses, flow cytometry) is an advantage.
- Relevant experience with the computational analysis of datasets using R is expected.
- Motivation to learn, apply and establish new techniques, ability to critically analyze and reflect data.
- Good skills with the following programs python, FlowJo, Adobe Illustrator, MS Office is an advantage.
- Good english language skills.
- Due to legal regulations, valid proof of measles immunity / measles vaccination is required.

The Department of Hematology / Oncology of the University Hospital Frankfurt offers a broad diagnostic and therapeutic spectrum for patients with solid tumors and malignant diseases of the hematopoietic system. The research team lead by Dr. Julius Enssle is dedicated to investigate translational research questions with systems biology methods in B cell neoplasms. Hereby, one focus is to decipher antiviral immunophenotyes in patients with Multiple Myeloma, the clonal trajectories of immune cell populations and their functional relevance (funded by the Else-Kröner-Fresenius-Foundation).This research is taking place within successful collaborations with the experimental immunology research group of Prof. Evelyn Ullrich (University Hospital Frankfurt), the stem cell research group of Prof. Michael Rieger (University Hospital Frankfurt) and the computational immunology group of Prof. Katharina Imkeller (UCT Frankfurt).
The research team itself is embedded in the excellent research infrastructure of the university hospital in Frankfurt. B cell lymphoma research and systems biology applications are one of the main cornerstones of the Department of Hematology / Oncology, as well as of the Frankfurt Cancer Institute and the DKTK partner site Frankfurt. Moreover there are multiple possibilities for collaborations with scientists from the Georg-Speyer-Haus, the cardiopulmonary institute as well as Robert-Koch-Institute (Berlin).
